 Detection of insulin receptor, phosphorylated on Tyr 1158, by immunoblotting. Samples: Whole cell lysate from CHO-T cells transfected with an insulin receptor containing vector and stimulated with insulin (lanes 1-4), or left unstimulated (lane 5). Primary antibody: PhosphoDetect™ Anti-IR/IGF1R (pTyr 1158) Rabbit pAb (Cat. No. 407706) (1:1000) pre-incubated without peptide (lanes 1, 5), with non-phosphorylated peptide corresponding to the immunogen (lane 2), a generic phosphotyrosine-containing peptide (lane 3), or phosphopeptide immunogen (lane 4). Detection: chemiluminescence.

Samples: CHO-T cells, serum-starved overnight, left untreated (left panel) or treated with insulin (100 nM, 10 min, right panel), fixed with 4% paraformaldehyde (20 min, RT), and permeabilized in PBS containing 0.1% saponin and 3% BSA (45 min, RT).
